Glyoxylic acid in the pharmaceutical industry

glyoxylic acid appearance

In the pharmaceutical industry, glyoxylic acid can be used as oral intermediates for penicillin, acetophenone, amino acids, α-thiophene glycolic acid, and p-hydroxyphenylacetamide.
Glyoxylic Acid Synthesis of P-hydroxymandelic Acid, p-Hydroxyphenylacetic Acid:

P-hydroxy mandelic acid was prepared by reacting phenol with glyoxylic acid under alkaline conditions, and then hydrogenated in a mineral acid or acetic acid aqueous solution to obtain p-hydroxyphenyl acetic acid, with a yield of about 70%. P-hydroxyphenylacetic acid is an important pesticide and pharmaceutical intermediate.
Synthesis of D-p-Hydroxyl Hydantoin from Glyoxylic Acid, D-P-Hydroxyphenylglycine:

The acid-catalyzed reaction of glyoxylic acid, phenol, and urea gives D-p-hydroxyphenylhydantoin, which is then hydrolyzed and converted directly by chemical resolution or enzymatic conversion of camphorsulfonic acid to obtain DN-carbamyl hydroxybenzene. Therefore, D-p-hydroxyphenylglycine was finally obtained by oxidizing with nitrous acid.
D-p-Hydroxyphenylglycine is mainly used in the manufacture of semi-synthetic antibiotics amoxicillin and semi-synthetic penicillins and cephalosporin drugs side chain compounds, can produce drugs amoxycillin, amoxycillin clavulanate, oxysphemine cephalosporin, Oxymetazole cephalosporin, cefoperazone, ceftriaxone, etc., are also used in the photographic field and as iron, phosphorus, silicon and other analytical reagents.
